DD ACE gene polymorphism is associated with increased coronary artery endothelial dysfunction: the PREFACE trial.

چکیده
Endothelial dysfunction is an early event in atherogenesis and is associated with the propensity to cause future cardiovascular events. The amelioration of endothelial dysfunction has been a research target for some years now, and success has been reported with, for example, the 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitors. Research has also identified the homozygous deletion (DD) subset of the angiotensin converting enzyme (ACE) insertion(I)/deletion(D) polymorphism to be associated with deteriorated endothelial function. The results, however, are conflicting. In a substudy of the PREFACE (pravastatin-related effects following angioplasty on coronary endothelium) trial we assessed ACE polymorphism, coronary endothelial function, and the influence of the HMG-CoA reductase inhibitor pravastatin.

Study of the correlation between ACE gene polymorphism and coronary artery disease
Introduction: Angiotensin converting enzyme (ACE) is an exopeptidase that converts Angiotensin I to Angiotensin II. Angiotensin II is a potent vasoconstrictor and releases aldosterone, and have a critical role in hypertension. In this study, ACE insertion / deletion (I/D) polymorphism and ACE activity was determined in patients with coronary artery disease (CAD) and normal subjects. The corr...
متن کاملMigraine and coronary artery disease: An updated open study on the genetic polymorphism
Genetic factors that increase susceptibility to oxidative stress, endothelial disfunction and, possibly, stroke include angiotensin-converting enzyme gene deletion polymorphism (ACE-DD) and the methylentetrahydropholate reductase (MTHFR) C677-TT polymorphism. The relationship of ACE-DD genotype to ischemic stroke and cardiovascular disease is controversial, but it has been independently linked ...
متن کاملDeletion polymorphism of the angiotensin I-converting enzyme gene in elderly patients with coronary heart disease.
Controversy exists as to whether the deletion/deletion (DD) genotype of angiotensin l-converting enzyme (ACE) gene polymorphism is associated with coronary heart disease (CHD). There are only a few studies dealing with this issue in the elderly, also with controversial results. The aim of this study was the assessment of correlation between genetic markers and the risk of CHD in the elderly. Th...
متن کاملAssociation between T-786C polymorphism of endothelial nitric oxide synthase gene and level of the vessel dilation factor in patients with coronary artery disease
Various polymorphisms on endothelial nitric oxide synthase (eNOs) gene cause reduced production of NO, the endothelial relaxing factor, and may accelerate the process of atherosclerosis. The study designed to investigate the frequency of T-786C polymorphism of the eNOs gene in patients suffering from coronary artery disease (CAD) in north-west of Iran. One hundred twenty subjects including 60 p...
متن کاملInsertion/deletion polymorphism of angiotensin-converting enzyme gene--risk factor for coronary artery disease in the Tuzla region population (Bosnia and Herzegovina).
Angiotensin II is the major effector molecule of renin-angiotensin system; its production can be conveniently interrupted by angiotensin-converting enzyme (ACE). Typical plasma levels of ACE accompany the I/D polymorphism; however, a controversy exists as to whether the DD genotype of the ACE polymorphism affects the risk for the development of coronary artery disease (CAD) and to what extent t...
